Dihydrolipoamide S-Acetyltransferase (DLAT) is one of three enzyme components of the pyruvate dehydrogenase complex. Specifically, DLAT catalyzes the transfer of an acetyl group to coenzyme A (CoA), which is the second step in the conversion of pyruvate into acetyl CoA during the tricarboxylic acid (TCA) cycle. Autoimmune reactivity against DLAT is the most common cause of primary biliary cholangitis, an autoimmune disease caused by autoantibodies against mitochondrial antigens. Additionally, mutations in this gene can cause pyruvate dehydrogenase E2 deficiency, resulting in primary lactic acidosis in infancy and early childhood. In line with the Warburg Hypothesis, which posits that disrupted cellular metabolism drives cancer formation, upregulation of DLAT is a characteristic of some cancers.

Recommended Usage: Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 1.25 - 5.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
Application Notes: This product is not recommended for use in IHC-P. This product does not recognize mouse DLAT in WB. When testing by ICC, the following fixation and permeabilization methods are suitable: ice-cold methanol, 4% PFA + 0.5% Triton-X, and 4%PFA + 100% methanol.
