TRIM29 (Tripartite Motif Containing 29), also known as ATDC (Ataxia-Telangiectasia Group D-Complementing), is a member of the TRIM protein family, which is characterized by a tripartite motif consisting of a RING finger domain, one or two B-box motifs, and a coiled-coil domain. Unlike most TRIM proteins, TRIM29 lacks the typical RING finger domain, which often confers E3 ubiquitin ligase activity. TRIM29 is primarily expressed in epithelial tissues such as the skin, gastrointestinal tract, and respiratory system. In normal physiological conditions, TRIM29 plays a crucial role in maintaining genomic stability and is involved in DNA repair pathways interacting with molecules such as p53-binding protein 1 (53BP1) and TIP60. Through these interactions, TRIM29 helps regulate DNA double-strand break (DSB) repair via the non-homologous end joining (NHEJ) and homologous recombination (HR) pathways. TRIM29 also functions in controlling epithelial cell differentiation and immune responses. TRIM29 can suppress type I interferon responses by regulating the STING pathway, which is essential for antiviral defense. In breast cancer and head and neck squamous cell carcinoma (HNSCC), TRIM29 often acts as an oncogene. Overexpression of TRIM29 in these cancers promotes tumor growth, invasion, and metastasis. Conversely, in gastric and colorectal cancers, TRIM29 act as a tumor suppressor. The expression is often reduced or silenced via promoter hypermethylation. The loss of TRIM29 function may lead to increased genomic instability and tumor progression.

It is recommended that the reagent be titrated for optimal performance for each application.
Additional Product Notes: For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704).
