The Platelet-derived growth factor (PDGF) family comprises of four different polypeptide chains encoded by four genes. The four PDGF chains assemble into disulfide-bonded dimers via homo- or heterodimerization. Five different dimeric isoforms have been described so far; PDGF-AA, PDGF-AB, PDGF-BB, PDGF-CC, and PDGF-DD. PDGF is a major mitogen for fibroblasts, smooth muscle cells, and others. They exert their functions by interacting with two related receptor tyrosine kinases, PDGF receptor α (PDGFRα) and PDGF receptor β (PDGFRβ). Upon ligand binding, PDGFRα undergoes dimerization and autophosphorylation, resulting in the formation of docking sites for SH2 domain-containing signaling proteins. These recruited molecules include enzymatic effectors (e.g., Src family kinases, phosphatidylinositol 3-kinase, Phospholipase Cy1, rasGAP, tyrosine phosphatase Syp) and adaptor proteins (e.g., GRB-2, Shc, Nck). The SH2 domains of these proteins specifically recognize phosphorylated PDGFRα, mediating the activation of downstream signaling pathways. PDGFRα plays a key role in many diseases through the above signal transduction.

Recommended Usage: Each lot of this antibody is quality control tested by blocking the effect of Recombinant Human PDGFRα-Fc Chimera (6 μg/mL) in NIH3T3 cells in a dose dependent manner. ND50 range: 1.5 – 10 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.
