Product Description

Bcl-X, a member of the Bcl-2 family of proteins, is encoded by the BCL2L1 gene and plays a crucial role in regulating apoptosis. Bcl-X exists in two main isoforms: Bcl-XL, which is anti-apoptotic, and Bcl-XS, which is pro-apoptotic. The balance between these isoforms is regulated through alternative splicing, influenced by various splice factors, transcription factors, and signaling pathways. Dysregulation of Bcl-X splicing has been implicated in several diseases, particularly cancer and diabetes. In cancer, overexpression of Bcl-XL can lead to resistance to chemotherapy, while in diabetes, increased Bcl-XS expression can result in apoptosis of pancreatic β-cells.

Recommended Usage: Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.125 - 1.0 µg/mL. For immunohistochemistry, a concentration range of 1.0 - 10.0 µg/mL is suggested. For flow cytometric staining, the suggested use of this reagent is ≤ 0.125 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
Application Notes: Due to homology, this antibody may detect the isoform Bcl-XS (18.8 kDa) which is a result of alternative splicing. However, in Western Blot, this isoform was not detected. The canonical sequence, isoform Bcl-XL, was detected by this antibody in Western Blot.
Additional Product Notes: For use of this antibody in intracellular flow cytometry (ICFC), we recommend fixation and permeabilization either using True-Nuclear™ Transcription Factor Buffer set (Cat. No. 424401) or Cyto-Fast™ Fix-Perm Buffer set (Cat. No. 426803). We DO NOT recommend using True-Phos™ Perm Buffer (Cat. No. 425401). For use of this antibody in immunohistochemistry on formalin-fixed paraffin-embedded tissues (IHC-P), we recommend antigen retrieval using either Citrate Buffer (Cat. No. 420902), or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (Cat. No. 422704).
