Heat shock protein B6 (HSPB6), also known as Hsp20, is a member of the small heat shock protein (sHSP) family. These proteins act as a molecular chaperone by stabilizing partially unfolded proteins, preventing aggregation, and assisting in refolding under stress conditions such as oxidative stress, ischemia, or heat shock. Hsp20 contributes to the maintenance of muscle relaxation and contractility. It interacts with cytoskeletal proteins like actin and myosin, and its phosphorylation, particularly at Ser16 by protein kinase A (PKA) or protein kinase G (PKG), enhances vasorelaxation by modulating actin dynamics and promoting smooth muscle relaxation. The phosphorylation of Hsp20 also confers protection against ischemia-reperfusion injury in cardiac tissue. Additionally, Hsp20 can inhibit apoptosis by interacting with pro-apoptotic proteins such as Bax and preventing mitochondrial membrane permeabilization, thereby maintaining mitochondrial integrity and cell survival. In cardiovascular disease, Hsp20 plays a cardioprotective role. Reduced expression or impaired phosphorylation of Hsp20 has been observed in failing hearts, correlating with increased susceptibility to myocardial infarction and heart failure. Overexpression of Hsp20 in animal models reduces infarct size and improves post-ischemic recovery, emphasizing its potential as a therapeutic target in ischemic heart disease.

Recommended Usage: Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunocytochemistry, a concentration range of 2.5 - 10.0 μg/mL is recommended. It is recommended that the reagent be titrated for optimal performance for each application.
Additional Product Notes: For use in immunocytochemistry (ICC), it is recommended to fix/permeabilize with the following: -Fixation buffer (Cat. No. 420801) with 0.5 % Triton X-100 For use in immunohistochemistry on formalin-fixed paraffin-embedded tissue (IHC-P), it is recommended to perform antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902) or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704).
