TNF-α is released from macrophages, monocytes, neutrophils, T-cells (principally CD4+), NK-cells and many transformed cell lines. Soluble homotrimer TNF-α is released from cells by proteolysis of the integral membrane precursor form of TNF-α. Binding of TNF-α to some of its receptors induces apoptosis.  Depending on cell types, receptor expression, and signal transduction status, this binding may induce other responses. In addition, TNF-α is involved in a wide range of pro-inflammatory response.

Recommended Usage: Each lot of this antibody is quality control tested by the neutralization of the cytotoxic effect induced by recombinant human TNF-α (Cat. No. 570104) at 3 ng/mL on L929 cells. ND50 range: 5 - 50 ng/mL. It is recommended that the reagent be titrated for optimal performance for each application.

However, we do provide references in which the antibodies were used for functional assays and we do perform QC to verify the specificity and quality of the antibody based on our strict specification criteria.
