Product Description

The c-MET protein, also known as the hepatocyte growth factor receptor (HGFR), is a receptor tyrosine kinase that plays a crucial role in various cellular processes. It is known to be involved in embryogenesis, tissue regeneration, and wound healing. c-MET is activated by binding to its ligand, hepatocyte growth factor (HGF), which triggers a cascade of intracellular signaling pathways such as PI3K/AKT, Ras/MAPK, and Wnt/β-catenin. Regulation of c-MET involves its interaction with other receptor tyrosine kinases like EGFR and RON, as well as its modulation by hypoxia-inducible factors. Aberrant activation of the c-MET/HGF axis, often due to gene mutations, overexpression, or amplification, is associated with the development and progression of various cancers in the liver, lung, and breast. This aberrant activation promotes tumor growth, angiogenesis, and metastasis, making c-MET a significant therapeutic target in oncology.

Application: WB - Quality tested ICC, ICFC - Verified
Recommended Usage: Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 2.5 - 10.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
Additional Product Notes: For use of this antibody in intracellular flow cytometry (ICFC), we recommend fixation and permeabilization using True-Phos™ Perm Buffer (Cat. No. 425401), True-Nuclear™ Transcription Factor Buffer set (Cat. No. 424401), or Cyto-Fast Fix/Perm Buffer set (Cat. No. 426803). For use of this antibody in Immunocytochemistry (ICC), we recommend fixation/permeabilization using PFA-Methanol, PFA-Triton X-100, or Methanol only.

Structure: The c-MET protein is composed of 1390 amino acids and has a molecular weight of approximately 155 kDa.
Distribution: Localized to cell surface as a receptor but may be found in the cytoplasm and nucleus under certain conditions. Expressed in hepatocytes, epithelial cells, and keratinocytes.
Function: • Acts as a receptor tyrosine kinase, binding to hepatocyte growth factor (HGF) to initiate signaling cascades. • Plays a crucial role in embryogenesis, tissue regeneration, and wound healing. • Involved in cell proliferation, motility, survival, and morphogenesis. • Promotes angiogenesis and invasive growth in cancer cells.
Interaction: • Hepatocyte Growth Factor (HGF): The primary ligand for c-MET, binding of HGF activates c-MET and triggers downstream signaling pathways such as PI3K/AKT, Ras/MAPK, and Wnt/β-catenin. • EGFR (Epidermal Growth Factor Receptor): c-MET can interact with EGFR, enhancing signaling pathways that promote cell proliferation, survival, and migration. • RON (Recepteur d'Origine Nantais): Another receptor tyrosine kinase that interacts with c-MET, contributing to the regulation of cell motility and invasive growth. • PI3K/AKT Pathway: c-MET activation leads to the stimulation of the PI3K/AKT pathway, which is crucial for cell survival and growth. • Ras/MAPK Pathway: This pathway is also activated by c-MET, playing a significant role in cell proliferation and differentiation. • SRC Family Kinases: c-MET interacts with SRC family kinases, which are involved in the regulation of cell adhesion, migration, and invasion
Ligand/Receptor: hepatocyte growth factor (HGF)
Cell Type: Epithelial cells
Biology Area: Cancer Biomarkers, Cell Biology, Stem Cells
