PIAS4 facilitates the attachment of SUMO (Small Ubiquitin-like Modifier) to target proteins and modulates protein stability, localization, and interactions. PIAS4 regulates the activity of transcription factors such as STATs (Signal Transducers and Activators of Transcription) by promoting their SUMOylation. PIAS4 participates in the DNA damage response, where it facilitates the recruitment of repair factors like 53BP1 to sites of DNA damage. This SUMOylation-driven process is vital for preserving genomic stability, preventing mutations, and reducing the risk of tumorigenesis. PIAS4 also influences chromatin dynamics by interacting with chromatin-associated proteins, affecting chromatin structure and gene expression patterns.
