Product Description

Insulin receptor substrate 1 (IRS1) is a crucial signaling adapter protein that mediates the effects of insulin and insulin-like growth factor 1 (IGF-1) by transmitting signals from their receptors to intracellular pathways such as PI3K/Akt and MAPK. The regulation of IRS1 involves phosphorylation at serine/threonine and tyrosine residues, which modulates its activity and stability. Dysregulation of IRS1 is implicated in several diseases, including type 2 diabetes mellitus and Alzheimer’s disease. In type 2 diabetes, defects in IRS1 signaling contribute to insulin resistance, while in Alzheimer’s disease, altered IRS1 signaling is associated with cognitive decline and neurodegeneration.

RRID: AB_3683497 (BioLegend Cat. No. 678351) AB_3683497 (BioLegend Cat. No. 678352)
Structure: Insulin receptor substrate 1 (IRS1) is a 1242 amino acid length protein with a molecular weight of 131.6 kDa. However, due to post-translational modifications like phosphorylation, it often appears as a 180 kDa protein when analyzed via SDS-PAGE.
Distribution: Insulin receptor substrate 1 (IRS1) is primarily located in the cytoplasm of the cell. However, upon activation by insulin or IGF-1, it is translocated to the plasma membrane where it interacts with the insulin receptor. It can also be found associated with the plasma membrane and other intracellular membrane-bound organelles as well as the nucleus.
Function: - Signal Transduction: Mediates insulin and IGF-1 signaling by transmitting signals from their receptors to intracellular pathways like PI3K/Akt and MAPK. - Metabolic Regulation: Plays a key role in glucose uptake, lipid metabolism, and overall energy homeostasis - Cell Growth and Survival: Involved in cellular processes such as growth, differentiation, and survival
Interaction: - Insulin Receptor (INSR): IRS1 binds to the insulin receptor via its pleckstrin homology (PH) domain and phosphotyrosine binding (PTB) domain, facilitating downstream signaling. - IGF-1 Receptor (IGF1R): IRS1 interacts with the IGF-1 receptor, promoting growth and metabolic effects. - PI3K (Phosphoinositide 3-kinase): IRS1 interacts with the regulatory subunit of PI3K (PIK3R1) through phosphorylated tyrosine residues, activating the PI3K/Akt pathway. - Grb2 (Growth factor receptor-bound protein 2): This interaction links IRS1 to the MAPK signaling pathway, influencing cell growth and differentiation. - SOCS7 (Suppressor of cytokine signaling - SOCS7 binds to IRS1 and can inhibit its signaling, acting as a negative regulator. - JAK2 (Janus kinase 2): IRS1 interacts with JAK2, which is involved in cytokine signaling and can modulate insulin signaling

Regulation: - Phosphorylation: Regulated by phosphorylation at serine/threonine and tyrosine residues, which affects its activity and stability - Feedback Inhibition: Negative feedback mechanisms, often involving serine phosphorylation, can inhibit IRS1 activity to prevent overactivation. - Protein-Protein Interactions: Interacts with various proteins that modulate its function and localization within the cell
