Aurora A (AURKA) is a serine/threonine kinase and a crucial regulator of mitosis. It is a member of the Aurora kinase family, which plays key roles in cell cycle regulation, spindle formation, and chromosome segregation. AURKA is highly conserved among eukaryotic organisms and is essential for maintaining genomic stability. Aurora A is predominantly active during the G2/M phase of the cell cycle which promotes the recruitment of pericentriolar material (PCM) to centrosomes, facilitating their maturation. Aurora A phosphorylates and activates several substrates involved in mitotic entry, such as PLK1 (Polo-like kinase 1) and CDC25B which enable the activation of cyclin-dependent kinases, driving the cell into mitosis. Aurora A stabilizes spindle microtubules by interacting with proteins such as TPX2 (Targeting Protein for Xklp2) and ensures proper bipolar spindle formation facilitating a prerequisite for accurate chromosome alignment and segregation. Aberrant expression or dysregulation of AURKA is implicated in oncogenesis. Overexpression is frequently observed in various cancers, including breast, ovarian, and colorectal cancers.

Recommended Usage: Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunocytochemistry, a concentration range of 5.0 - 10.0 μg/mL is recommended. For flow cytometric staining, the suggested use of this reagent is ≤ 0.25 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
Application Notes: For use in immunocytochemistry (ICC), it is recommended to fix/permeabilize with either of the following: -Fixation buffer (Cat. No. 420801) with 0.5 % Triton X-100 -Fixation buffer (Cat. No. 420801) with 100% ice-cold methanol For use in intracellular flow cytometry (ICFC), it is recommended to fix/permeabilize with True-Nuclear™ Transcription Factor Buffer Set (Cat. No. 424401). Fixation/permeabilization with True-Phos™ Perm Buffer (Cat. No. 425401) or Cyto-Fast™ Fix/Perm Buffer Set (Cat. No. 426803) is not recommended.
