L-Type Amino Acid Transporter 1 (LAT1) is crucial for transporting large neutral amino acids like leucine, isoleucine, and phenylalanine across cell membranes, playing a significant role in cellular metabolism and nutrient supply. LAT1 is highly expressed in the blood-brain barrier, where it facilitates the delivery of essential amino acids to the brain. Dysregulation of LAT1 is associated with several diseases, including cancer, where it supports tumor growth by supplying amino acids necessary for rapid cell proliferation. Recommended Usage: Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.25 - 1.0 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 1.0 - 10.0 µg/mL is suggested. For immunohistochemistry, a concentration range of 1.25 - 10.0 µg/mL is suggested. For flow cytometric staining, the suggested use of this reagent is ≤ 0.06 µg per million cells in 100 µL volume. It is recommended that the reagent be titrated for optimal performance for each application.
Application Notes: For use of this antibody in Immunocytochemistry (ICC), we recommend fixation/permeabilization using either PFA-Methanol or Methanol only. We do not recommend PFA-Triton X-100 for fixation/permeabilization.For use of this antibody in immunohistochemistry on formalin-fixed paraffin-embedded tissues (IHC-P), we recommend antigen retrieval using either Citrate Buffer, 10X (Cat. No. 420902), or Tris-EDTA pH 9.0 Antigen Retrieval Buffer (10X) (Cat. No. 422704).For use of this antibody in intracellular Flow Cytometry (ICFC), we recommend fixation/permeabilization using True-Phos™ Perm Buffer (Cat. No. 425401).
