Interleukin-24 (IL-24), a cytokine belonging to the IL-10 family, is produced by activated monocytes, macrophages, T cells, keratinocytes, and epithelial cells. Mature mouse IL-24 shares approximately 69% amino acid sequence identity with human IL-24. IL-24 exerts its biological effects by signaling through two heterodimeric receptor complexes, IL-20R1/IL-20R2 or IL-22R1/IL-20R2, leading to the activation of the JAK/STAT, MAPK, and PI3K/AKT pathways. Functionally, IL-24 plays a critical role in immune regulation, host defense, inflammation, cell proliferation, differentiation, wound healing, and tissue remodeling. In monocytes, IL-24 induces the production of proinflammatory cytokines such as TNF-α, IL-6, and IL-1β, while in B cells, it suppresses plasma cell differentiation. IL-24 selectively induces apoptosis in various cancer cell types, including melanoma, breast, lung, prostate, and pancreatic cancers. IL-24 also suppresses angiogenesis by inhibiting endothelial cell proliferation and migration. Furthermore, IL-24 enhances the recruitment and activation of immune cells, such as NK cells and cytotoxic T lymphocytes, within the tumor microenvironment. While IL-24 contributes to host defense mechanisms, it can also promote pathological inflammation observed in conditions such as rheumatoid arthritis, psoriasis, and inflammatory bowel disease.

Molecular Mass: The 167 amino acid recombinant protein has a predicted molecular mass of approximately 19.4 kD. The DTT-reduced and non-reduced proteins migrate at approximately 19-23 kD by SDS-PAGE. The N-terminal amino acid is Met.

Storage & Handling: Unopened vial can be stored between 2°C and 8°C for up to 2 weeks, at -20°C for up to six months, or at -70°C or colder until the expiration date. For maximum results, quick spin vial prior to opening. The protein can be aliquoted and stored at -20°C or colder. Stock solutions can also be prepared at 50 - 100 µg/mL in appropriate sterile buffer, carrier protein such as 0.2 - 1% BSA or HSA can be added when preparing the stock solution. Aliquots can be stored between 2°C and 8°C for up to one week and stored at -20°C or colder for up to 3 months. Avoid repeated freeze/thaw cycles.
Activity: ED50 = 1 – 12 ug/mL as measured by its ability to support the adhesion of COLO 205 cells.

A: The specific activity range of the protein is indicated on the product datasheets. Because the exact activity values on a per unit basis can largely fluctuate depending on a number of factors, including the nature of the assay, cell density, age of cells/passage number, culture media used, and end user technique, the specific activity is best defined as a range and we guarantee the specific activity of all our lots will be within the range indicated on the datasheet. Please note this only applies to recombinants labeled for use in bioassays. ELISA standard recombinant proteins are not recommended for bioassay usage as they are not tested for these applications.
Q: Have your recombinants been tested for stability?
A: Our testing shows that the recombinant proteins are able to withstand room temperature for a week without losing activity. In addition the recombinant proteins were also found to withstand four cycles of freeze and thaw without losing activity.
