The complement system is a part of eukaryotic innate defense system that enhances the clearance of exogenous microbes and damaged cells by antibodies and phagocytic cells. In vivo , the complement system is activated by three pathways: the classical complement pathway, the alternative complement pathway, and the lectin pathway. Complement C5 is a 1676 amino acid precursor protein synthesized in the liver. All three complement activation pathways culminate in the cleavage of C5, which produces two disulfide linked glycoproteins C5α and C5β. C5α is further cleaved by C5 convertase into C5a and C5α', the latter binds to C5β to form C5b, which in turn sequentially recruits C6-C9 to form the membrane attack complex (MAC). MAC destroys pathogens by forming a lytic pore in their membrane. Mutations affecting the Complement C5 gene lead to complement component 5 deficiency where patients demonstrate an increased propensity to develop severe recurrent infections.

Recommended Usage: Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 2.0 - 10 µg/mL. For immunohistochemistry on formalin-fixed paraffin-embedded tissue sections, a concentration range of 2.0 - 10 µg/mL is suggested. It is recommended that the reagent be titrated for optimal performance for each application.
Application Notes: This antibody predominantly detects C5b in human cell line and human plasma. At high concentration (between 2 and 10 µg/mL), the full-length C5 is also detected at ~190 kD in human cell lines, but not in human plasma.
