Casein Kinase 1 Alpha 1 (CK1α) is a member of the CK1 family of serine/threonine kinases which phosphorylate various regulatory molecules. CK1α is one of seven different non-allelic casein kinase isoforms, each with distinct signaling partners conferred by the sequence deviation in their C-terminal extensions. It plays a critical role in Wnt/β-catenin signaling through phosphorylation of β-catenin at serine 45 by GSK-3, which primes β-catenin for additional phosphorylation events, and is a critical component of the Axin-GSK-3-β-catenin scaffold. Through its phosphorylation of tumor suppressor p53 at serine 20, CK1α plays an important role in cell cycle regulation, DNA repair, apoptosis, and tumor suppression, thus making it of particular interest as a therapeutic target. Studies suggest that CK1α interacts with keratin-associated protein FAM83H to mediate the organization of keratin filaments and cytoskeletal organization.

Recommended Usage: Each lot of this antibody is quality control tested by western blotting. For western blotting, the suggested use of this reagent is 0.125 - 1.0 µg/mL. It is recommended that the reagent be titrated for optimal performance for each application.
Application Notes: This clone was tested for ICC using HeLa cells fixed with 4% paraformaldehyde and permeabilized with either methanol or Triton X-100. Neither method resulted in CK1a staining. This clone was tested for IP using whole cell extracts prepared from HeLa cells. The clone failed to immunoprecipitate CK1a. This clone recognizes multiple isoforms of CK1a.
