Background ETV5 (ERM) is a member of the PEA3 (polyomavirus enhancer activator 3) subfamily of the E26 transformation-specific (ETS) DNA-binding transcription factors (1,2). PEA3 subfamily members are considered as oncogenic transcriptional factors due to their close association with and contribution to cancer development. The subfamily of ETS proteins consists of three members: ETV1, ETV4, and ETV5. RAS/MAPK and PKA are identified to be the upstream activator of ETV5 by phosphorylation of the target at serine residues (3-5). In addition to cancer promoting effect, ETV5 also regulates many basic cellular functions during normal development, including embryonic stem cell differentiation (6,7), neuronal cell growth, differentiation, and neuron-dendrite network establishment (8,9), reproduction organ development (10,11), as well as cellular fatty acid metabolism and obesity (12,13).
