Background
Deleted in colorectal cancer (DCC) is a type I transmembrane protein belonging to the immunoglobulin superfamily. Originally implicated as a tumor suppressor gene given its lack of expression in colorectal carcinomas, DCC was later characterized as a receptor for netrin (1). Netrin proteins belong to an evolutionarily conserved family of laminin-like molecules that play an essential role in axon guidance during development. These secreted proteins can have opposing functions depending on specific receptor associations. Binding of netrin-1 by DCC on commissural axons mediates growth cone attraction, while binding by uncoordinated 5 (UNC5) family receptors induces repulsion (2,3). In addition to axon guidance, DCC is also suggested to regulate tissue organization, cell adhesion, motility, proliferation, differentiation, and cell survival via its binding activity with netrin-1 (1,3). Recent findings suggest that reduced expression of netrin-1 or DCC can lead to dopaminergic (DA) neuron death within the substantia nigra pars compacta (SNpc) of the midbrain, an area that is particularly vulnerable to degeneration in Parkinson's disease (PD). Conversely, overexpression of netrin-1 in animal models of PD has been shown to be neuroprotective, suggesting that the netrin-1/DCC ligand/receptor pair may be a therapeutic target for this neurodegenerative disease (4,5).
