Background
Glutamate dehydrogenase 1 and 2 (GluD1 and GluD2, respectively), also known as glutamate receptor ionotropic, delta-1 and delta-2 (GRID1 and GRID2, respectively), are part of the delta family of ionotropic glutamate receptors (iGluRs). Ionotropic receptors, typically cation-permeable ligand-gated ion channels, mediate most excitatory transmission in the brain (1). iGluR classes are defined based on sequence identity and pharmacological properties and include α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptors, kainate receptors, and N-methyl-D-aspartate (NMDA) receptors (2). Although GluDs were identified based on sequence identity and their structure is related to other iGluRs, GluDs do not bind glutamate and are unlikely to form ion channels like other iGluRs (3). GluD1 and GluD2 share 56% sequence homology. While GluD2 expression is highest in the cerebellum and likely regulates motor function, GluD1 is expressed throughout the forebrain. While GluDs may not function like typical ligand-gated ion channels, they bind to other synaptic proteins and may function to alter subcellular expression of synaptic proteins, including other iGluRs as well as synaptic adhesion proteins. Genetic studies in mice suggest GluDs function in higher-order functions, including learning, memory, and addiction.
