Background
Cathepsin Z, also known as cathepsin X or cathepsin P, is a lysosomal cysteine proteinase member of the papain family encoded by the gene (1,2). It contains a uniquely short pro-domain containing an RGD integrin binding sequence, allowing functionality in cell adhesion, signaling, and extracellular matrix remodeling (3,4). Cathepsin Z is expressed in immune cells, specifically macrophages, monocytes, and dendritic cells, where it participates in several immune cell functions (5). It may play a role in tumorigenesis and has been implicated in several cancer types, including hepatocellular carcinoma, glioblastoma, non-small cell lung cancer, prostate cancer, and colorectal cancer (6-10). Cathepsin Z is also expressed and secreted by activated microglia in the brain and may play a role in neuroinflammation (11,12).
