Background N-Myc and STAT interactor (NMI) is an interferon inducible protein that functions as a transcriptional regulator (1). Research studies have demonstrated that intracellular pools of NMI, in complex with interferon-induced 35 kDa protein (IFI35), negatively regulate innate immune antiviral signaling by blunting type I interferon production (2,3). NMI has been shown to interact with a variety of transcription factors that play a role in tumorigenesis, including N-myc, c-Myc, BRCA1, STAT1, STAT3, STAT5, and NF-κB (4-8). Its downregulation has been linked to enhanced tumor growth and invasiveness in lung and breast cancers, respectively (9,10). Conversely, elevated NMI expression in glioblastoma correlates with poor prognosis, suggesting that NMI's role in cancer is tumor-dependent (11).
