Product Description
Monoclonal Antibody for studying PPP1R14B. Validated for Western Blotting,Immunoprecipitation,Immunohistochemistry (Paraffin). Available in 2 sizes. Highly specific and rigorously validated in-house, PPP1R14B/PHI-1 (F6O6F) Rabbit Monoclonal Antibody (CST #25729) is ready to ship. Product Usage Information Western Blotting: 1:1000 Immunoprecipitation: 1:100 Immunohistochemistry (Paraffin): 1:400 - 1:800 Storage Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at -20°C. Do not aliquot the antibody. Protocol Available protocols: Western Blotting, Immunoprecipitation, Immunohistochemistry (Paraffin) Specificity / Sensitivity PPP1R14B/PHI-1 (F6O6F) Rabbit Monoclonal Antibody recognizes endogenous levels of total PPP1R14B/PHI-1 protein. This antibody does not cross-react with PPP1R14A/CPI-17. Non-specific staining was observed in mouse brain, spleen, and liver by immunohistochemistry. Species Reactivity: Human, Mouse, Rat, Monkey Source / Purification Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Val50 of human PPP1R14B/PHI-1 protein. Background Phosphatase holoenzyme inhibitor (PHI-1), encoded by the protein phosphatase 1 (PP1) regulatory subunit 14B gene ( ), belongs to a family of proteins, including PPP1R14A/CPI-17, PPP1R14C/KEPI, and PPP1R14D/GEPI that each specifically inhibits a subset of PP1 holoenzymes. PHI-1 is highly conserved among eukaryotes and ubiquitously expressed at low levels across tissue types, but it is overexpressed in a wide array of cancer types, including colon and lung adenocarcinomas (1,2). PHI-1 also displays a proteostatic function, both directly and indirectly, on several targets in the MAPK signaling pathways. In hepatocellular carcinoma (HCC) cells, PHI-1 bound to, stabilized, and increased phosphorylation of p90RSK at Ser363/Ser380, and PHI-1 knockdown inhibited HCC cell growth (3). In HEK 293 cells, PHI-1 knockdown stabilized c-Raf protein and increased downstream ERK1/ERK2 activation (4). PHI-1 phosphorylation at Thr57 increases its inhibitory activity toward PP1 50-fold, and it has been established as a PKC and ILK substrate (5,6). Alternate Names PHI-1; phospholipase C beta-3 neighboring gene protein; Phospholipase C-beta-3 neighbouring gene protein; PLCB3N; PNG; PP14B; PPP1R14B; protein phosphatase 1 regulatory inhibitor subunit 14B; Protein phosphatase 1 regulatory subunit 14B; protein phosphatase 1, regulatory (inhibitor) subunit 14B; SOM172 Specification REACTIVITY: H M R Mk SENSITIVITY: Endogenous MW (kDa): 19 Source/Isotype: Rabbit IgG
