Product Usage Information
Prior to use, thaw the recombinant protein and briefly spin down to collect all liquid at the bottom of the tube. Dilute according to the following instructions depending on the ELISA kit type. An example sensitivity curve is shown in Figure 2. PathScan ® ELISA Kits: Dilute with ELISA Sample Diluent to achieve a 1X protein concentration. PathScan ® RP ELISA Kits: Dilute with 1X Cell Lysis Buffer #9803 to a 2X protein concentration to achieve a final 1X protein concentration upon addition of the detection antibody. FastScan™ ELISA Kits: Dilute with 1X FastScan™ ELISA Cell Extraction Buffer to a 2X protein concentration to achieve a final 1X protein concentration upon addition of the antibody cocktail.

Expression Host: 293 Supplied in 1X PBS (8.1mM Na 2 HPO 4 , 2.68 mM KCl, 1.675 mM KH 2 PO 4 , and 137 mM NaCl (pH 7.4)).

Background
The poly-Glycine-Serine (G4S) linker is a type of flexible, unstructured synthetic peptide linker sequence often leveraged to connect the variable heavy (VH) domain and variable light (VL) domain of single-chain variable fragments (scFvs) and chimeric antigen receptors (CARs) that utilize an extracellular domain scFv for target antigen recognition. The linker itself consists of a core pentapeptide sequence, Gly-Gly-Gly-Gly-Ser, that is repeated and commonly found as either a 15-mer (G4S) or 20-mer (G4S) within scFv-based CARs and scFv fragments. The linker sequence length plays a role in controlling scFv stability and the noncovalent association between the VH and VL domains (1,2).
