Background LZTFL1 (leucine zipper transcription factor-like 1) is highly expressed in the epithelial cells of normal tissues and is a negative regulator of BBSome ciliary trafficking and sonic hedgehog (SHH) signaling. The absence of LZTFL1 leads to a phenotype with enhanced developmental abnormalities, and mutations in LZTFL1 have been reported in patients with Bardet-Biedl syndrome (BBS) (1,2). LZTFL1 has been shown to suppress epithelial mesenchymal transition (EMT), and decreased LZTFL1 expression is associated with tumor growth, metastasis, and poor survival outcome in many cancers (3-5). Genome-wide association studies have identified a gain of function allele in an enhancer region that leads to increased expression of LZTFL1, which is functionally associated with more severe outcomes from SARS-CoV-2 infection (6).
