Product Usage Information For optimal ChIP results, use 10 μL of antibody and 10 μg of chromatin (approximately 4 × 10 6 cells) per IP. This antibody has been validated using SimpleChIP ® Enzymatic Chromatin IP Kits. Western Blotting: 1:1000 Immunoprecipitation: 1:50 Chromatin IP: 1:50 Storage Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at -20°C. Do not aliquot the antibody. Background MPP8, also known as MPHOSPH8, is a methylated H3K9 reader protein that interacts with histone methyltransferases ESET and GLP (1). The N-terminal chromodomain specifically recognizes di- and tri-methyl H3K9 over methylated H3K27 and can be phosphorylated, contributing to its binding activity (2,3). MPP8 forms a dimer that serves as a bridge between G9a, GLP, and DNMT3A to set up cytosine methylation and H3K9 tri-methylation at target loci (4). MPP8 is a member of the human silencing hub (HUSH) complex along with TASOR, PPHLN1, and MORC2, which localizes to H3K9me3 sites and cooperates with SETDB1 to maintain H3K9me3 levels and transcriptional repression (5). The HUSH complex is capable of silencing long interspersed elements (LINEs) located in euchromatic regions (6). The HUSH complex can also play a role in the viral life cycle by silencing transcript production from viral genomes (7).
