Background Activating transcription factor 7-interacting protein (ATF7IP), also known as MCAF1 or AM, is a repressive chromatin factor that binds to SETDB1, enhancing its ability to catalyze histone H3 lysine 9 trimethylation (1-3). ATF7IP has a SUMO binding domain, which interacts with sumoylated MBD1 to recruit the SETDB1/ATF7IP heterodimer to sites of heterochromatin (4). Loss of ATF7IP results in degradation of SETDB1 protein and a global loss of H3K9me3 (5). ATF7IP can localize to PML bodies in a SUMO-dependent manner, causing reduced histone mRNA and protein and cellular senescence (6). ATF7IP has also been implicated in cancer, where it interacts with SP1 to regulate telomerase. It can also contribute to immune cell function by inhibiting the and genes (7-9).
