Background
Mitochondrial glycine amidinotransferase (GATM), also known as arginine:glycine amidinotransferase (AGAT), catalyzes the conversion of glycine and L-arginine to guanidinoacetate and L-ornithine, the rate-limiting step of creatine biosynthesis (1-3). Mouse model studies of obesity-accelerated breast cancer show that this enzyme and creatine biosynthesis in adipocytes are essential to breast cancer progression driven by obesity (2). In addition, synthesis of creatine by GATM or dietary uptake of creatine promotes tumor metastasis in an orthotopic mouse model (3). Moreover, GATM expression increases in metastases of various cancers, such as colorectal cancer and melanoma (3). Furthermore, creatine plays a role in neurons and its function requires GATM and SLC6A8, a creatine transporter (4).
