Background Vitamin K epoxide reductase complex subunit 1-like protein 1 (VKORC1L1) catalyzes the reduction of vitamin K to vitamin K hydroquinone, a radical-trapping antioxidant. Studies show that VKORC1L1 is a potent ferroptosis suppressor. It protects cells from ferroptosis independent of the GSH/GPX4 pathway by generating vitamin K hydroquinone, counteracting lipid peroxidation. In addition, is a direct transcriptional repression target of p53. p53 activation inhibits VKORC1L1 expression, sensitizing cells to ferroptosis for tumor suppression. Furthermore, it was shown that the VKORC1L1 inhibitor warfarin promotes ferroptosis and thus suppresses tumor growth in pancreatic cancer mouse models (1,2).
