Background Ribosomal protein L15 (RPL15) is a structural component of the large 60S subunit of the ribosome. Studies show that overexpressed RPL15 protein promotes metastatic growth of breast cancer patient-derived circulating tumor cells. Overexpression of RPL15 was shown to increase translation of other ribosomal proteins of large and small subunits and cell cycle regulators (1). Furthermore, the DNA topoisomerase I inhibitor topotecan binds to RPL15 and suppresses preribosomal subunit formation, which causes ribosomal stress and enhances DNA topoisomerase I-independent antitumor immune activation mediated by damage-associated molecular patterns (2). In addition, RPL15 expression is upregulated in colon cancer cells. RPL15 deficiency leads to ribosomal stress and, therefore, apoptosis of colon cancer cells (3).
