Product Description
Monoclonal Antibody for studying PTPN2 iso2. Validated for Western Blotting. Highly specific and rigorously validated in-house, TCPTP (TC45) (D7T7D) Rabbit Monoclonal Antibody (BSA and Azide Free) (CST #29857) is ready to ship. Product Usage Information This product is the carrier free version of product #58935. All data were generated using the same antibody clone in the standard formulation which contains BSA and glycerol. This formulation is ideal for use with technologies requiring specialized or custom antibody labeling, including fluorophores, metals, lanthanides, and oligonucleotides. It is not recommended for ChIP, ChIP-seq, CUT&RUN or CUT&Tag assays. If you require a carrier free formulation for chromatin profiling, please contact us . Optimal dilutions/concentrations should be determined by the end user. BSA and Azide Free antibodies are quality control tested by size exclusion chromatography (SEC) to determine antibody integrity. Formulation Supplied in 1X PBS (10 mM Na 2 HPO 4 , 3 mM KCl, 2 mM KH 2 PO 4 , and 140 mM NaCl (pH 7.8)). BSA and Azide Free. For standard formulation of this product see product # 58935 Storage Store at -20°C. This product will freeze at -20°C so it is recommended to aliquot into single-use vials to avoid multiple freeze/thaw cycles. A slight precipitate may be present and can be dissolved by gently vortexing. This will not interfere with antibody performance. Specificity / Sensitivity TCPTP (TC45) (D7T7D) Rabbit Monoclonal Antibody (BSA and Azide Free) recognizes endogenous levels of total TCPTP (TC45) protein. This antibody does not recognize the 48 kDa (TC48) isoform. Species Reactivity: Human, Mouse, Rat, Hamster, Monkey Source / Purification Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Pro382 of human TCPTP (TC45) protein. Background T cell protein tyrosine phosphatase (TCPTP, PTPN2, PTN2) is a non-receptor protein tyrosine phosphatase (PTP) that regulates signal transduction pathways by catalyzing the dephosphorylation of tyrosine residues (1). Two described TCPTP splice variants include a 48 kDa isoform (TC48) that is targeted to secretory pathway organelles (e.g., endoplasmic reticulum) by a hydrophobic carboxy terminus, and a 45 kDa isoform (TC45) that actively shuttles between the nucleus and cytoplasm (2). TCPTP substrates include receptor and non-receptor tyrosine kinases, such as EGFR, JAK1/3, and Src-family kinases, as well as STAT3 and other nuclear substrates (3-6). Research studies show that the corresponding gene is deleted in a subset of human T-cell acute lymphoblastic leukemias. The loss of TCPTP has been suggested to promote tumor progression through enhanced JAK/STAT signaling (7,8).
