PALB2 acts as a molecular scaffold for the BRCA1/BRCA2/RAD51 DNA repair complex, and functions to help recruit this complex to sites of DNA damage and stimulate RAD51 DNA strand-invasion activity required for D-loop formation (1). PALB2 is a tumor suppressor protein that is required for homologous repair of double-stranded DNA breaks. Mutations in PALB2 (mostly mono-allelic truncations) lead to increased susceptibility to breast, prostate, and pancreatic cancers (2-4).
