Protocol
Available protocols: Western Blotting, Immunoprecipitation, Immunofluorescence (Immunocytochemistry)
Specificity / Sensitivity
Liprin alpha1 (E7D7W) Rabbit Monoclonal Antibody recognizes endogenous levels of total Liprin α1 protein. A band of unknown origin at 78 kDa is detected by western blot. No specific signal was observed in fixed frozen mouse tissue by immunofluorescence.
Species Reactivity: Human
Source / Purification
Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Pro819 of human Liprin α1 protein.
Background
The Liprin α family contains four members (Liprin α1/2/3/4) in vertebrates, which are core synaptic scaffolding proteins essential for synapse assembly and maturation (1). Liprin α proteins form homodimers and heterodimers with Liprin β proteins, and all four genes encoding Liprin α family members undergo alternative splicing in humans and mice (2,3). Liprin α1 is ubiquitously expressed and regulated in an activity-dependent manner. Liprin α1 is degraded either in response to CaMKII phosphorylation or via the E3 ubiquitin ligase anaphase-promoting complex (APC) (4). At the synapse, it participates in both presynaptic and postsynaptic functions, including active zone assembly, neurotransmitter release, and synaptic cargo transport, as well as acetylcholine receptor (AChR) clustering and organization of postsynaptic machinery (1,2). In squamous cell carcinoma of head and neck (HNSCC) cell lines, Liprin α1 has been shown to regulate biological processes related to protein trafficking, cell-cell contacts, and cell-substrate contacts. Liprin α1 also affects the distribution of cell surface β1 integrin, thereby promoting focal adhesion turnover and controlling cell edge dynamics, supporting its additional role in cancer cell invasion and metastasis pathways (5,6).Â
