Background
The E26 transformation-specific (ETS) transcription factors bind to a core 5'-GGAA/T-3' DNA motif and are known to regulate many biological processes, including cell proliferation, differentiation, apoptosis, hematopoiesis, and angiogenesis; they have also been described in pathological conditions, including cancer (reviewed in 1). ETS transcription factor E74-like factor 3 (ELF3), also called ESE-1, ESX, Jen, and ERT, belongs to a subtype of ETS proteins characterized by a predominant epithelial-specific expression profile (2-6). ELF3 shows widespread expression across tissues with high expression in the digestive tract. Targeted knockout of the gene in mice results in lethality in about 30% of the embryos to E11.5, along with abnormalities in the intestinal epithelium (7). ELF3 can have dualistic roles in cancer with tumor-promoting and suppressing activity (reviewed in 8,9). Dysregulation of ELF3 through genetic and epigenetic mechanisms such as inactivating mutations, promoter methylation, and gene amplification have been reported across multiple cancer types and stages of disease. In addition, ELF3 has been shown to play a role in immune disorders and inflammation (10-12).
