Product Usage Information Western Blotting: 1:1000 Simple Western™: 1:10 - 1:50 Storage Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Species Reactivity: Human, Mouse, Rat Source / Purification Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human DEGS1 protein. Background Sphingolipid delta(4)-desaturase (DES1, DEGS1), also known as dihydroceramide desaturase-1, inserts a double bond into the sphingoid backbone of ceramides in most tissues. Studies show that deletion of from mice deficient in leptin improves glucose homeostasis and resolves insulin resistance and hepatic steatosis. These findings indicate that ceramides contribute to metabolic disorders (1). In addition, DEGS1 is involved in hematopoietic stem cell (HSC) function. In culture, before quiescence exit, inhibition of DEGS1 in human HSCs by its inhibitor 4HPR activates stress response pathways, including the unfolded protein response (UPR) and autophagy, to maintain HSC self-renewal (2). Furthermore, DEGS1 is in the mitochondria-associated endoplasmic reticulum membrane (MAM) and is critical to MAM functions (3).
