Background Guanylate-binding proteins (GBPs) are a family of dynamin-related proteins that facilitate host defense against intracellular bacterial and viral pathogens (1). Interferon (IFN) signaling upregulates the expression of several GBPs in response to infection, including the most extensively studied paralog GBP1 (1). GBP1 specifically binds to lipopolysaccharide (LPS) on the cell surface of cytosolic Gram-negative bacteria, forming a stable protein layer and disrupting the integrity of the outer membrane (2). GBP1 also recruits caspase-4 to the membrane and promotes the pro-inflammatory cell death program known as pyroptosis (3,4). GBP1 is increasingly studied in the context of cancer, where it appears to restrict cellular proliferation in breast and colorectal cancers but may promote growth and chemoresistance in cases of ovarian cancer and glioblastoma (5-9). Elevated GBP1 expression levels also correlate with enhanced immune cell infiltration and increased overall survival in breast cancer patients treated with immunotherapy (10).
