Background Muscle-specific RING finger protein 1 (MuRF1) is an E3 ubiquitin ligase that promotes muscle cell protein degradation during muscle atrophy (1). In response to conditions that promote muscle atrophy, elevated skeletal muscle MuRF1 facilitates the ubiquitination and degradation of thick filament components (2). Cardiac muscle MuRF1 promotes cardiomyocyte troponin I degradation and inhibits PKCε-mediated cardiac hypertrophy (3,4). MuRF1 also regulates degradation of the large sarcomeric protein titin, the transcriptional regulator GMEB1, the SUMO conjugating enzyme UBC9, and the ubiquitin peptidase USP13 (5). Binding of FoxO1 and the glucocorticoid receptor to adjacent sites within the MuRF1 promoter region activates transcription of this E3 ubiquitin ligase (6).
