Background Triosephosphate isomerase (TPI1) catalyzes the interconversion of dihydroxyacetone phosphate (DHAP) and glyceraldehyde-3-phosphate (GAP). TPI1 expression levels were shown to increase in lung adenocarcinomas and inversely correlate with overall patient survival (1). In addition, mTORC1 phosphorylates and activates cyclin-dependent kinase 2 (CDK2). Activated CDK2, in turn, phosphorylates TPI1 at Ser117, stimulating TPI1 to translocate to the nucleus. Nuclear TPI1 reduces DHAP levels, leading to acetate accumulation in the nucleus and, therefore, impacting histone acetylation (2). Furthermore, the salt-inducible kinases (SIKs), which are activated by the serine/threonine protein kinase LKB1, phosphorylate TPI1 at Ser21 to regulate its activity in human lung adenocarcinomas (3).
