Product Description
Monoclonal Antibody for studying hnRNP U. Validated for Western Blotting. Available in 2 sizes. Highly specific and rigorously validated in-house, hnRNP U (E2O1R) Rabbit Monoclonal Antibody (CST #34095) is ready to ship. Product Usage Information Western Blotting: 1:1000 Storage Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/ml BSA, 50% glycerol and less than 0.02% sodium azide. Store at -20°C. Do not aliquot the antibody. Protocol Available protocols: Western Blotting Specificity / Sensitivity hnRNP U (E2O1R) Rabbit Monoclonal Antibody recognizes endogenous levels of total hnRNP U protein. Species Reactivity: Human, Mouse, Rat, Monkey Source / Purification Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Val479 of human hnRNP U protein. Background hnRNP U, also known as scaffold attachment factor A (SAF-A) or SP120, was first described as a nuclear matrix protein that contributes to RNA metabolism (1-3). hnRNP U has been shown to bind pre-mRNAs and contributes to splicing by controlling U2 snRNP maturation (4). Furthermore, hnRNP U has been shown to bind chromatin to help regulate its structure during interphase (5). During prophase, hnRNP U becomes phosphorylated by Aurora-B, which evicts hnRNP U and the chromatin-associated RNAs bound to it, a process critical to proper chromosome alignment (6,7). hnRNP U plays a role in the chromatin localization of Xist RNA, and loss of hnRNP U in ES cells fail to achieve an inactivated X chromosome (8). hnRNP U has also been shown to play a key role in immunity, functioning as a viral dsRNA sensor. Upon viral recognition, hnRNP U oligomerizes and activates numerous antiviral genes, including IFNB1 (9). Loss of function variants of hnRNP U have been shown to cause early onset epilepsy and intellectual disorders (10,11).
