Background NSUN5 was originally discovered as a yeast 5-methylcytosine methyltransferase that methylates C2278 of the 25S ribosomal RNA (rRNA) (1). It also methylates C3782 in human 28S rRNA, and contributes to protein synthesis and cell growth (2). NSUN5 is overexpressed in many cancer types, where it contributes to increased cellular proliferation (3,4). NSUN5 also plays an important role in cerebral cortex development, and NSUN5 mutants have neurological dysfunction (5-7).
