Background Ras GTPase-activating protein 2 (RASA2) belongs to the GAP1 subfamily of Ras-GAPs and serves as one of the major Ras-specific GAPs. RASA2 functions to inhibit Ras by facilitating activation of its intrinsic GTPase activity. Similar to other members of the GAP1 subfamily, RASA2 contains calcium-binding C2 domains and a phospholipid binding PH domain (1). Research studies have pointed to RASA2 as an inhibitor of Ras signaling in the context of human cancer, including melanoma (2). More recently, RASA2 has been identified as a negative regulator of T cell effector function through its function as a Ras-specific GAP (3,4).
