Background
GTP cyclohydrolase 1 (GCH1) catalyzes the rate-limiting step of tetrahydrobiopterin (BH ) biosynthesis. Studies show that BH functions as a radical-trapping antioxidant and protects cells against ferroptosis independently of GPX4. In addition, dihydrofolate reductase (DHFR) is required to maintain BH levels by regenerating the compound (1,2). Furthermore, deficiency of spliceosomal GTPase Eftud2 in mouse cerebellar Purkinje cells (PCs) causes severe ferroptosis, PC degeneration, dyskinesia, and cerebellar atrophy. Ablation of in PCs downregulates expression through mis-splicing of , activating ferroptosis (3).
