Background Pre-mRNA-processing factor 8 (PRPF8) is the largest protein component of the human small nuclear ribonucleoprotein (snRNP) U5 and interacts with U2 and U12-dependent spliceosomes (1,2). PRPF8 forms a structure that creates a cavity for the spliceosome's catalytic center and binds to two other critical U5 components, SNRNP200 and EFTUD2. PRPF8, along with EFTUD2, affects the function of SNRNP200, an RNA helicase that unwinds the U4/U6 heterogeneous nuclear ribonucleoprotein (hnRNP) duplex to activate the spliceosome (1,3-6). snRNP U5 assembly is facilitated by HSP90 and R2TP chaperones (7). Viral RNA polymerase has been found to target PRPF8 to inhibit pre-mRNA splicing and mRNA synthesis (8).
