Product Description
Monoclonal Antibody for studying MLH1. Validated for Western Blotting,Immunofluorescence (Immunocytochemistry),Flow Cytometry (Fixed/Permeabilized). Highly specific and rigorously validated in-house, MLH1 (4C9C7) Mouse Monoclonal Antibody (BSA and Azide Free) (CST #37715) is ready to ship. Product Usage Information This product is the carrier free version of product #3515. All data were generated using the same antibody clone in the standard formulation which contains BSA and glycerol. This formulation is ideal for use with technologies requiring specialized or custom antibody labeling, including fluorophores, metals, lanthanides, and oligonucleotides. It is not recommended for ChIP, ChIP-seq, CUT&RUN or CUT&Tag assays. If you require a carrier free formulation for chromatin profiling, please contact us . Optimal dilutions/concentrations should be determined by the end user. BSA and Azide Free antibodies are quality control tested by size exclusion chromatography (SEC) to determine antibody integrity. Formulation Supplied in 1X PBS (10 mM Na 2 HPO 4 , 3 mM KCl, 2 mM KH 2 PO 4 , and 140 mM NaCl (pH 7.8)). BSA and Azide Free. For standard formulation of this product see product # 3515 Storage Store at -20°C. This product will freeze at -20°C so it is recommended to aliquot into single-use vials to avoid multiple freeze/thaw cycles. A slight precipitate may be present and can be dissolved by gently vortexing. This will not interfere with antibody performance. Specificity / Sensitivity MLH1 (4C9C7) Mouse Monoclonal Antibody (BSA and Azide Free) detects endogenous levels of total MLH1 protein. Species Reactivity: Human, Monkey Source / Purification Monoclonal antibody is produced by immunizing animals with truncated recombinant MBP-MLH1. Background Mismatch repair (MMR), a conserved process that involves correcting errors made during DNA synthesis, is crucial to the maintenance of genomic integrity. MLH1 is the human homologue of the MMR gene . MMR requires recognition of a base mismatch or insertion/deletion loop by a MutS homolog followed by recruitment of a MutL heterodimeric complex consisting of MLH1 and PMS1 (MutL-γ), PMS2 (MutL-α), or MLH3 (MutL-γ). Other factors required for MMR in eukaryotes are EXO1, PCNA, RFC, RPA, DNA polymerases, and DNA ligases (reviewed in 1). Inactivation of the gene causes genome instability and predisposition to cancer (2-5). The gene is frequently mutated in hereditary nonpolyposis colon cancer (HNPCC) (6). MLH1 also plays a role in meiotic recombination (7). Alternate Names COCA2; DNA mismatch repair protein Mlh1; FCC2; hMLH1; HNPCC; HNPCC2; MGC5172; MLH1; mutL homolog 1; mutL homolog 1, colon cancer, nonpolyposis type 2; mutL homolog 1, colon cancer, nonpolyposis type 2 (E. coli); MutL protein homolog 1 Specification REACTIVITY: H Mk SENSITIVITY: Endogenous MW (kDa): 85 Source/Isotype: Mouse IgG1
