Background Interferon-gamma-inducible GTPase 10 (IRGB10) is a member of a family of interferon-inducible GTPases, also known as p47 GTPases (1). In mice, there are 23 genes present in the p47 GTPase family; only two are found in human, neither of which are orthologs of mouse (2,3). In macrophages, IRGB10 expression is upregulated by IRF1 in response to infection by pathogenic bacteria, such as and (4). Along with guanylate binding proteins (GBPs), IRGB10 binds to and disrupts bacterial cell membranes, exposing bacterial ligands such as lipopolysaccharide (LPS) and bacterial DNA (4). These ligands may then be recognized by sensors such as AIM2 and caspase-11, leading to inflammasome formation (4). Similarly, IRGB10 targets the fungal cell wall of during infection to combat the pathogen (5). Interestingly, unlike in the context of bacterial and fungal infection, in which IRGB10 contributes to host defense, in the context of influenza A infection, IRGB10 facilitates viral replication; mice were more resistant to influenza A infection than wild-type mice (6).
