Background The leukemia-associated Rho guanine nucleotide exchange factor (LARG, also known as ARHGEF12) was originally identified as a novel fusion partner of the mixed-lineage leukemia protein in an acute myeloid leukemia patient. LARG activates RhoA and RhoC but not Rac1 or Cdc42. (1,2). It has been shown to play a role in glioblastoma cell invasion and resistance (2). Furthermore, LARG is associated with certain forms of synaptic plasticity, where, as a binding partner of GluD1, it plays a role in inhibitory synapse formation (3).
