Background NLR family pyrin domain containing 12 (NLRP12, NALP12, PYPAF7, MONARCH-1), a member of the NOD-like receptor (NLR) family, is an intracellular protein that plays a role in forming an inflammasome and can also act as a regulator of inflammatory signaling (1). NLRP12 is expressed in dendritic cells, neutrophils, granulocytes, and macrophages (2). Like other NLR family members, NLRP12 is composed of a pyrin domain (PYD), a nucleotide-binding domain (NBD), and a leucine-rich repeat (LRR) domain (2). NLRP12 functions as a negative regulator of canonical NFκB signaling by inhibiting accumulation of hyperphosphorylated IRAK1 downstream of TLR signaling (3). NLRP12 also acts as a negative regulator of non-canonical NFκB signaling by associating with NFκB-inducing kinase (NIK) and TRAF3, which leads to proteasomal degradation of NIK (4, 5). During Yersinia pestis infection, NLRP12 acts as an inflammasome component, resulting in the release of IL-1β and IL-18 (6). NLRP12 plays a role in regulating gut microbiota, reducing susceptibility to colitis (7).
