Background Autism susceptibility candidate 2 (AUTS2) is a transcriptional activator protein that was first identified in a pair of homozygous twins with autism spectrum disorder (ASD) (1). AUTS2 associates with the non-canonical polycomb repressive complex 1 (ncPRC1) and recruits the histone acetyltransferase p300 to activate gene expression (2). AUTS2 is primarily expressed in early developing brain tissue and contributes to neuronal differentiation (3). AUTS2 is expressed as either a full-length "long" protein isoform or a "short" isoform lacking the N-terminus due to an alternative transcription start site (4). The exact biological differences between the two isoforms are not fully understood, but they may target distinct genes during differentiation (5).
