Species Reactivity: Human, Mouse, Rat Source / Purification Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues near the carboxy terminus of human UBASH3A protein. Background Ubiquitin-associated and SH3 domain-containing protein A (UBASH3A), also referred to as Suppressor of T cell receptor signaling 2 (STS-2), is an atypical protein tyrosine phosphatase that is primarily expressed in T cells (1). UBASH3A is a modular protein that contains an N-terminal UBA domain that can interact with ubiquitin, a central SH3 domain that mediates protein-protein interactions, and a C-terminal histidine phosphatase domain (1,2). Although it displays limited phosphatase activity compared to its homolog UBASH3B, UBASH3A functions as a negative regulator of T cell activation by downregulating the TCR-CD3 complex in a phosphatase-independent manner (3). UBASH3A also associates with the E3 ubiquitin ligase Cbl-b, which further dampens T cell activation through downregulation of CD28 (3). Due to its role in regulating T cell responses, UBASH3A dysfunction has been linked to several autoimmune disorders, including type 1 diabetes, rheumatoid arthritis, celiac disease, and systemic lupus erythematosus (4-7).
