Background Spi-C belongs to the erythroblast transformation-specific (ETS) domain family of transcription factors and binds to the purine-rich PU-box (1-3). Spi-C is a lineage-instructive factor involved in generating mature, specialized subsets of both myeloid and lymphoid cells (6). Spi-C is highly expressed by red pulp macrophages in the spleen, which are required for red blood cell recycling and iron homeostasis (4,6). Spi-C is also transiently expressed in B cells during specific stages of their development and regulates cell fate decisions at the pro-B to pre-B cell stage and plasma cell differentiation (1-3,6). Spi-C has also been shown to be a positive regulator that promotes osteoclast differentiation and function (5).
