Background
Nicotinic acetylcholine receptors (nAChRs) are ligand-gated ion channels found throughout the peripheral and central nervous system (CNS), involved in fast synaptic transmission and a wide range of physiological functions (1). In mammals, neuronal nAChRs are classified through combinations of 11 alpha (α2-α7, α9, and α10) and beta (β2-β4) subunits, with the α7 and α4β2 subtypes being the most prevalent in the brain (2). Dysfunction of these receptors has been implicated in several neurological diseases, including Alzheimer's disease (AD), Parkinson's disease (PD), schizophrenia, and neuropathic pain (3-5). Transmembrane protein 35 (TMEM35), also called novel acetylcholine receptor chaperone (NACHO), is a molecular chaperone encoded by the gene. It is crucial for the proper assembly, surface expression, and function of several neuronal nAChR subtypes in the brain, especially the α7 subtype (6). CNS α7 nAChRs have been strongly implicated in AD and PD, making TMEM35 a potential therapeutic target and tool to better understand CNS nAChRs in neurodegeneration (3-5). TMEM35 has also been linked to the development and progression of osteosarcoma (OSA) by playing a role in OSA cell proliferation and migration (7).
