Background Nuclear speckles (NS) are distinct biomolecular condensates consisting of nuclear proteins and RNA molecules. Although their exact biological function is not fully understood, NS are believed to function as hubs for mRNA splicing and potentially transcription (1). The splicing factors serine/arginine repetitive matrix protein 2 (SRRM2) and SON are necessary for NS formation (2). SRRM2 and SON undergo phase separation and function as scaffolding proteins, recruiting additional splicing factors and promoting NS assembly (3). SON has been shown to be critical for maintaining pluripotency in human embryonic stem cells (hESCs) by splicing key regulatory gene transcripts (4).
