Background DNA ligase III/LIG3 is an ATP-dependent DNA ligase that functions in DNA repair. Four isoforms of LIG3 result from alternative splicing/initiation, including a mitochondrial-specific isoform essential for cell viability (1). Loss of nuclear LIG3 sensitizes resistant cells to PARP inhibitors and enhances PARP inhibitor efficacy in BRCA1-deficient mouse mammary tumors (2). In multiple myeloma (MM), LIG3 is crucial for repair of DNA double-stranded breaks (DSBs) via alternative-non-homologous end joining (Alt-NHEJ), and LIG3 inhibitors are being investigated as cancer therapeutics (3).
